
A specific anti-phospho-peptide antibody (Ab) labeled with an europium chelate (Eu) is used in conjunction with a peptide substrate directly labeled with the new ULight acceptor dye for the detection of kinase activity. The binding of the Eu Ab to the phosphorylated ULight peptide substrate brings both the donor and acceptor dye molecules into close proximity. Upon irradiation at 320/340 nm, the excited Eu transfers its energy to the nearby ULight dye molecule, which in turn emits light at 665 nm. The intensity of light emission is proportional to the level of phosphorylation. |
