Deoxynucleotides for Indirect Detection

NEN brand nucleotide analogs are biologically active with a variety of DNA and/or RNA polymerases. Labeling methods such as nick translation, random priming, polymerase chain reaction, 3’-end labeling, or transcription of RNA using SP6, T3, or T7 RNA polymerases may be used. For best results, we DO NOT RECOMMEND using these analogs at 100% level of substitution in the labeling reaction. Labeled probes may be used in applications including (but not limited to) Southerns, Northerns, colony blots and chromosome mapping. Hapten labeled analogs are detected by indirect methods. Indirect detection is mediated via enzyme labeled streptavidin or antibody directed against the specific moiety. The antibody labeled with an enzyme such as horseradish peroxidase or alkaline phosphatase is detected using chromogenic or chemiluminescence reagents. Signal amplification may be obtained using NEN’s patented Tyramide Signal Amplification process (TSA™). TSA amplified signals may be either colorimetric, chemiluminescent, or fluorescent.
